Cancer antibodies are the economically most successful class of biologics with 2007 sales of 15.7 bln for antibodies targeting CD20, VEGF, Her2 and EGF-R. No wonder that nobody wanted to miss the train of the new (old) antibody target insulin-like growth factor-1 receptor (IGF-1R). Meanwhile, eight different antibodies are in clinical development and further two are close to. Not enough, at least four small molecules are in clinical evaluation. The most favourite, but rather rare clinical indication Ewing sarcoma (osteosarcoma) is studied by at least five companies. The most obvious differentiation characteristic of the antibodies is their frequence of administration, i.e. once weekly or once every two or three weeks. Whether other smarter biochemical differences play a clinical role, remains to be seen when clinical results of advanced studies will be released.
